Overview
BGM-2121 is a humanized IgG1 monoclonal antibody developed by BioGate Precision Medicine Corp., a member of the Powerchip Group. It specifically targets the N-terminal region (amino acids 1-34) of parathyroid hormone-related peptide (PTHrP), a protein frequently overexpressed in various cancers that contributes to tumor growth, humoral hypercalcemia of malignancy (HHM), and cancer cachexia. By neutralizing PTHrP, BGM-2121 blocks the PTHrP/PTH1R signaling pathway. Preclinical studies have demonstrated significant antitumor activity, including the reduction of tumor growth and the reversal of hypercalcemia and weight loss in models of lung squamous cell carcinoma, pancreatic cancer, and colorectal cancer. The drug is currently undergoing Phase 1 clinical evaluation to assess its safety, pharmacokinetics, and preliminary efficacy in patients with advanced solid tumors who have exhausted standard treatment options.
