BGN siRNA is a small interfering RNA therapeutic candidate designed to silence the expression of the Biglycan (BGN) gene. Biglycan is a small leucine-rich proteoglycan encoded on the X chromosome that has been identified as a key driver of sex differences in pulmonary arterial hypertension (PAH). Research conducted at UCLA indicates that BGN is significantly upregulated in the right ventricles and lungs of PAH patients, particularly females, where it promotes a proliferative, fibrotic, and anti-apoptotic phenotype in pulmonary artery smooth muscle cells (PASMC) and fibroblasts (PAFB). By silencing BGN, the siRNA aims to attenuate pulmonary artery vascular remodeling and reduce pulmonary hypertension. In preclinical models, intratracheal administration of BGN siRNA has shown efficacy in reducing monocrotaline-induced pulmonary hypertension in female rats.
