BH-30643 + carboplatin + pemetrexed

BH-30643 + carboplatin + pemetrexed is an investigational combination regimen being developed by BlossomHill Therapeutics for the treatment of locally advanced or metastatic non-small cell lung cancer (NSCLC) harboring EGFR and/or HER2 mutations. The regimen combines BH-30643, a first-in-class, orally available, macrocyclic, non-covalent, CNS-active, and mutant-selective small molecule OMNI-EGFR™ inhibitor, with the standard-of-care chemotherapy agents carboplatin and pemetrexed. BH-30643 is designed to target a broad range of EGFR mutations, including classical, atypical (PACC), and resistance mutations like C797S, while sparing wild-type EGFR and HER2 to reduce off-target toxicities. The combination is currently being evaluated in the Phase 1/2 SOLARA trial (NCT06706076) to determine the recommended Phase 2 dose and assess preliminary antitumor activity and safety in patients with advanced NSCLC.
