BH1621 is an albumin-bound nanoparticle formulation of docetaxel developed by Zhuhai Beihai Biotech for the treatment of various solid tumors. As a next-generation taxane, it utilizes human serum albumin as a carrier to deliver docetaxel, a cytotoxic agent that binds to and stabilizes microtubules, thereby inhibiting mitotic division and inducing apoptosis in cancer cells. This "nab" (nanoparticle albumin-bound) technology is designed to enhance the solubility of docetaxel without the need for toxic solvents like polysorbate 80 (Tween 80), which are associated with severe hypersensitivity reactions and require corticosteroid premedication. BH1621 leverages albumin-mediated transport mechanisms, such as binding to the gp60 receptor and subsequent transcytosis, to potentially increase intratumoral drug concentration and improve the overall safety and efficacy profile compared to conventional docetaxel formulations. It is currently undergoing clinical evaluation in Phase 1 and Phase 2 trials for indications including breast cancer, non-small cell lung cancer, and gastric cancer.
