BH3120 is a heterodimeric, IgG-like bispecific antibody developed using Hanmi Pharmaceutical's proprietary Pentambody platform. It is designed to simultaneously target programmed death-ligand 1 (PD-L1) on tumor cells and 4-1BB (CD137) on immune cells. By blocking PD-L1, BH3120 inhibits the immune checkpoint pathway that tumors use to evade immune detection, while its agonistic action on 4-1BB provides co-stimulatory signals to T cells within the tumor microenvironment (TME). This dual mechanism aims to enhance anti-tumor immunity specifically in tumor tissues while minimizing systemic immune activation and associated toxicities. Preclinical studies have demonstrated strong antitumor efficacy with minimal off-target effects, and early clinical trials are evaluating its safety and efficacy as monotherapy or in combination with PD-1 antagonists for advanced or metastatic solid tumors[1][2][3][5][6].
