BHB810 is a novel, site-specific antibody-drug conjugate (ADC) targeting Cadherin-17 (CDH17), developed by BigHat Biosciences using its AI/ML-powered Milliner platform. The molecule comprises a humanized single-domain VHH-Fc fusion antibody conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE) via a serum-stable, enzyme-cleavable linker (GlycoConnect SYNstatin E technology). BHB810 is engineered for high serum stability and superior tissue penetration due to its smaller size (approximately 50% of a standard IgG). Preclinical data demonstrate robust internalization and potent anti-tumor activity in gastric cancer models, including those with low or heterogeneous CDH17 expression. First-in-human clinical trials are anticipated to begin in 2026, focusing on advanced gastric and other gastrointestinal malignancies.
