BHQ880 placebo is the inactive comparator used in clinical trials evaluating BHQ880, a fully human IgG1 monoclonal antibody developed by Novartis. BHQ880 is designed to neutralize Dickkopf-1 (DKK1), a soluble inhibitor of the Wnt signaling pathway that is overexpressed in multiple myeloma and contributes to osteolytic bone disease by suppressing osteoblast function. The placebo was utilized in a Phase 2 randomized, double-blind study (NCT01337752) where it was administered in combination with bortezomib and dexamethasone to patients with previously untreated multiple myeloma and renal insufficiency. The primary purpose of the placebo arm in this context is to provide a baseline for assessing the efficacy of BHQ880 in increasing bone mass and preventing skeletal-related events (SREs).
