BHV-1310 is a novel, bispecific small molecule degrader of immunoglobulin G (IgG), developed by Biohaven for the treatment of autoimmune diseases, with an initial focus on myasthenia gravis. It is engineered to rapidly and selectively lower circulating IgG levels via asialoglycoprotein receptor (ASGPR)-mediated hepatic lysosomal degradation. This mechanism enables deep and fast onset of IgG-lowering, with a short time to maximal effect and brief exposure period. Notably, BHV-1310 is designed to spare IgG3—preserving immune effector function—while targeting other subclasses (IgG1, IgG2, IgG4), potentially reducing the risk of broad immunosuppression compared to other therapies. Preclinical studies in animal models have demonstrated rapid (>90% within 24 hours), robust, safe, and selective reduction in circulating IgG without affecting other immunoglobulin isotypes or key clinical parameters such as albumin or cholesterol. The drug can be co-administered with biologics due to its selectivity and lack of interference with Fc-containing therapeutics[1][3][5][6].
