BHV-1510 is a next-generation, fully optimized antibody-drug conjugate (ADC) targeting trophoblast cell surface antigen 2 (Trop-2), developed for the treatment of advanced or metastatic epithelial tumors. The drug consists of a Trop-2-directed antibody conjugated to a proprietary topoisomerase 1 (TopoIx) payload via a homogeneous drug-antibody ratio (DAR) of four and employs unique site-specific conjugation with a highly stable and irreversible linker designed by GeneQuantum Healthcare. Preclinical studies have demonstrated that BHV-1510 induces cellular cytotoxicity, bystander killing, and immunogenic cell death, leading to enhanced monotherapy efficacy and synergistic effects when combined with anti-PD1 therapy. The agent has shown superior efficacy and safety compared to other Trop-2 ADCs in development, notably lacking lung toxicity in preclinical models. Biohaven is conducting Phase 1/2 clinical trials as both monotherapy and in combination with cemiplimab (Libtayo), an anti-PD1 monoclonal antibody supplied by Regeneron[1][3][4][5][6][8].
