Overview

BI 1358894 is a novel small molecule inhibitor of the transient receptor potential cation channel subfamily C members 4 and 5 (TRPC4/5). It is being developed primarily for neuropsychiatric indications including major depressive disorder, borderline personality disorder, and post-traumatic stress disorder. The drug acts by inhibiting TRPC4 and TRPC5 ion channels, which are implicated in neuronal signaling pathways relevant to mood regulation and emotional processing. Clinical trials have shown that while the primary efficacy endpoints were not met in some studies (e.g., borderline personality disorder), BI 1358894 was generally well tolerated with a safety profile comparable to placebo[1][6]. The compound originated from Hydra Biosciences and is being co-developed by Boehringer Ingelheim.
