BI 1387446 is a small molecule STING (stimulator of interferon genes) agonist developed by Boehringer Ingelheim for the treatment of advanced solid tumors. It is administered via intratumoral injection and works by binding to and activating the STING pathway in immune cells within the tumor microenvironment, leading to increased production of type I interferons and other cytokines that promote anti-tumor immune responses. The drug has demonstrated dose-dependent pharmacodynamic effects, including upregulation of IFN pathway genes such as CXCL10 and IFIT1, as well as increased plasma levels of IP-10 (CXCL10) and IFNγ following administration. As of April 2024, BI 1387446 has completed phase I clinical trials in patients with advanced or metastatic solid tumors[1][2][3][4][5][6][7].
