Overview

BI-1622 is a small molecule inhibitor of Human Epidermal Growth Factor Receptor 2 (HER2), originally developed by Boehringer Ingelheim for the treatment of HER2-positive cancers. Recent research has explored its potential for repurposing in the treatment of Type 1 and Type 2 diabetes. The drug works by inhibiting maladaptive HER2 activation in pancreatic beta-cells under pro-diabetic, gluco-lipotoxic, and pro-inflammatory conditions, which otherwise contributes to beta-cell dysfunction, stress signaling, and loss of identity. In preclinical models, BI-1622 has demonstrated the ability to restore normoglycemia, improve insulin secretion, and promote the survival and maturation of beta-cells.
