BI 1703880 is a synthetic, second-generation STING (Stimulator of Interferon Genes) agonist developed for the treatment of advanced solid tumors. It works by activating the STING pathway, an intracellular nucleic acid sensor that plays a key role in innate immunity. Activation of STING induces a type I interferon response and upregulates immune-related proteins such as PD-L1 and human leukocyte antigen in tumor cells, thereby enhancing antitumor immune activity. Preclinical studies have demonstrated antitumor effects, and early clinical trials are evaluating its safety and efficacy both as monotherapy and in combination with ezabenlimab (an anti-PD-1 antibody). The drug is administered intravenously to patients with advanced cancers who have failed standard treatments or for whom no standard treatment exists[1][4][6][7].
