Overview

**BI 1831169 + anti-PD-1 antibody** is an investigational combination therapy under clinical evaluation for advanced solid tumors. **BI 1831169** (also known as VSV-GP) is a genetically modified, live, recombinant oncolytic virus based on vesicular stomatitis virus (VSV), in which its native neurotropic glycoprotein G is replaced with the glycoprotein (GP) of lymphocytic choriomeningitis virus, reducing neurotoxicity. The oncolytic virus is designed to selectively infect, replicate in, and lyse cancer cells and stimulate antitumor immune responses in an interferon-dependent manner. **Anti-PD-1 antibody** in the combination is ezabenlimab (BI 754091), an immune checkpoint inhibitor designed to block the programmed death-1 (PD-1) pathway, thereby enhancing T-cell mediated antitumor immunity. The combination aims to synergize direct viral oncolysis and immune activation (by BI 1831169) with inhibition of immune suppression (by anti-PD-1 antibody), potentially resulting in improved antitumor efficacy. The therapy is under investigation in first-in-human, dose-escalation trials for unresectable, refractory, or metastatic solid tumors[4][9].
