Overview

BI 3819026 is a bispecific T-cell engager (TcE) antibody designed to target Glypican-3 (GPC3) and the CD3 epsilon subunit of the T-cell receptor complex. Developed by Boehringer Ingelheim, the molecule is engineered to bridge GPC3-expressing tumor cells with cytotoxic T-cells, thereby inducing T-cell activation, proliferation, and the release of cytolytic proteins like perforin and granzymes to eliminate the cancer cells. GPC3 is an oncofetal antigen that is highly overexpressed in several malignancies, most notably hepatocellular carcinoma (HCC), while remaining largely absent in healthy adult tissues, making it a promising target for precision immunotherapy. BI 3819026 is currently undergoing Phase 1 clinical evaluation as both a monotherapy and in combination with the PD-1 inhibitor ezabenlimab (BI 754091) in patients with advanced or metastatic solid tumors.
