Overview

BI 3820768 is an investigational bispecific T-cell engager (TcE) developed by Boehringer Ingelheim for the treatment of solid tumors. It is designed to bind simultaneously to Claudin 6 (CLDN6), a tight junction protein frequently overexpressed in specific cancers such as germ cell tumors, ovarian cancer, and endometrial cancer, and the CD3 subunit of the T-cell receptor complex. By bridging these two targets, BI 3820768 facilitates the formation of an immunological synapse, leading to T-cell activation, cytokine release, and the targeted lysis of CLDN6-expressing malignant cells. As of 2024, it is being evaluated in a first-in-human Phase I clinical trial to assess its safety, tolerability, and preliminary efficacy in patients with advanced relapsed or refractory tumors.
