BI 3822971

BI 3822971 is a bispecific T-cell engager (TCE) developed by Boehringer Ingelheim for the treatment of advanced solid tumors. The molecule is designed to simultaneously target B7-H3 (CD276), a checkpoint molecule frequently overexpressed on the surface of various cancer cells and tumor-associated vasculature, and the CD3 epsilon subunit of the T-cell receptor complex. By cross-linking B7-H3-positive tumor cells with cytotoxic T cells, BI 3822971 triggers T-cell activation, secretion of cytotoxic cytokines, and directed lysis of the tumor cells. As of 2024, BI 3822971 is undergoing Phase 1 first-in-human clinical evaluation to assess its safety, tolerability, and pharmacokinetics in patients with advanced malignancies.
