Overview

BI 3923948 is an investigational oncolytic virus therapy developed by Boehringer Ingelheim for the treatment of advanced, unresectable, or metastatic solid tumors. It utilizes the VSV-GP platform, which is a chimeric vesicular stomatitis virus (VSV) where the native glycoprotein has been replaced with the glycoprotein of the lymphocytic choriomeningitis virus (LCMV). This modification is designed to enhance the safety profile by reducing neurotropism and to allow for repeated dosing by avoiding rapid neutralization by the host immune system. BI 3923948 is administered via intratumoral injection, where it selectively replicates in and lyses cancer cells, releasing tumor-associated antigens and inducing a pro-inflammatory environment to trigger a systemic anti-cancer immune response. It is currently being evaluated in Phase I clinical trials as a monotherapy and in combination with the PD-1 inhibitor ezabenlimab.
