BI 414 is a potent, orally available small molecule antagonist of the melanin-concentrating hormone receptor 1 (MCHR1). It was developed by Boehringer Ingelheim as part of their MCHR1 antagonist program targeting obesity. The compound belongs to the biphenylacetamide chemical class and was optimized for improved potency, selectivity over related GPCRs, and brain exposure. Preclinical studies demonstrated that BI 414 has acceptable in vivo efficacy in animal models of obesity[1][2].
