Overview

BI-749327 is a potent, highly selective, and orally bioavailable small molecule antagonist of the **Transient receptor potential cation channel subfamily C member 6 (TRPC6)**, developed by Boehringer Ingelheim. It demonstrates high selectivity for TRPC6 over its close homologs TRPC3 and TRPC7. Preclinical studies indicate that BI-749327 can ameliorate cardiac and renal fibrosis, improve left heart function in pressure overload models, and may have applications in treating Duchenne muscular dystrophy (DMD). Furthermore, recent research suggests its role in inhibiting breast cancer metastasis by suppressing TRPC6-mediated resistance to ferroptosis, achieved through the repression of c-Myc and maintenance of glutathione levels.
