Overview

**BI 765063 + BI 770371** is an investigational combination of two humanized monoclonal antibodies targeting the SIRPα (signal regulatory protein alpha) pathway, developed for immuno-oncology indications. Both drugs antagonize SIRPα, a receptor that, when bound to CD47 on tumor cells, delivers a "don’t eat me" signal to myeloid cells, preventing effective anti-tumor immunity. BI 765063 specifically blocks SIRPα to activate macrophage-mediated tumor destruction. BI 770371, a next-generation IgG1 monoclonal antibody, recognizes both V1 and V2 SIRPα alleles, making it suitable for a broader range of cancer patients. The combination aims to disrupt CD47/SIRPα signaling and restore myeloid cell anti-tumor function. These drugs are being investigated for advanced solid tumors and head and neck squamous cell carcinoma, both as monotherapy and in combination with other immune checkpoint inhibitors, including ezabenlimab (an anti-PD-1 antibody) and cetuximab[1][2][3][6][10].
