Overview

**BI 770371 + ezabenlimab** is a combination of two investigational monoclonal antibodies developed for immuno-oncology. **BI 770371** is a next-generation IgG1 monoclonal antibody targeting SIRPα (signal regulatory protein alpha), designed to bind both V1 and V2 allelic variants, thereby blocking the interaction between SIRPα and CD47 ("don’t eat me" signal) to enhance innate immune responses by restoring the function of myeloid cells in the tumor microenvironment. **Ezabenlimab** (BI 754091) is a programmed death-1 (PD-1) immune checkpoint inhibitor antibody. The combination aims to synergistically activate both innate (via SIRPα antagonism) and adaptive (via PD-1 blockade) immunity to induce anti-tumor responses. The regimen is under clinical investigation primarily for advanced solid tumors, including head and neck squamous cell carcinoma, with emerging exploratory interest in metabolic diseases such as MASH (metabolic dysfunction-associated steatohepatitis)[2][7][10][1][4][8].
