BI 811283 is a small molecule inhibitor that selectively targets Aurora B kinase, a serine/threonine-protein kinase involved in the regulation of mitosis. By binding to and inhibiting Aurora B kinase, BI 811283 disrupts the assembly of the mitotic spindle apparatus, impairs chromosome segregation, and inhibits cell proliferation. This mechanism leads to growth inhibition and senescence in cancer cells. Preclinical studies have shown potent antitumor activity across various cancer models, including non-small cell lung cancer (NSCLC), colorectal cancer, pancreatic cancer, and others. In clinical trials (phases I/II), BI 811283 demonstrated disease stabilization in some patients with advanced solid tumors or acute myeloid leukemia (AML). The most common adverse effects are hematological toxicities such as neutropenia and leukopenia[1][2][6][8].
