Overview

**BI 831266** is a potent, selective, low-molecular-weight small molecule inhibitor targeting **Aurora kinase B**. Aurora kinase B is a serine/threonine kinase crucial for mitosis, and its inhibition results in impaired cell division, leading to cell cycle arrest and apoptosis in tumor cells. BI 831266 was under investigation for the treatment of advanced solid tumors, including colorectal, cervical, and other cancers. Preclinical studies demonstrated antiproliferative activity in multiple cancer cell lines. In phase 1 clinical trials, BI 831266 was administered intravenously but the studies were terminated early for reasons unrelated to efficacy or safety, and the maximum tolerated dose was not established. The compound was largely developed for oncology indications and showed manageable toxicity with some evidence of antitumor response[1][3][5][6][9].
