BI 836826 is a chimeric immunoglobulin G1 (IgG1) monoclonal antibody that targets human CD37, a transmembrane protein highly expressed on mature B lymphocytes and malignant B cells. It is engineered to enhance binding to human Fcγ receptors, thereby improving its ability to induce antibody-dependent cell-mediated cytotoxicity (ADCC) against CD37-overexpressing tumor cells. The drug also induces apoptosis in these tumor cells. Preclinical studies demonstrated potent depletion of normal and malignant B cells, as well as significant antitumor activity in lymphoma models. Developed using XmAb technology and cytotoxic Fc domain engineering, it was investigated for the treatment of relapsed or refractory chronic lymphocytic leukemia (CLL), diffuse large B-cell lymphoma (DLBCL), non-Hodgkin's lymphoma (NHL), follicular lymphoma, and unspecified respiratory disorders. Clinical development for hematologic malignancies was discontinued after phase I trials[1][3][4][5][7].
