BI 894416 is a novel, potent, and highly selective small molecule inhibitor of spleen tyrosine kinase (SYK), developed by Boehringer Ingelheim for the treatment of severe asthma. The drug was designed to modulate immune responses implicated in respiratory diseases by targeting SYK, a key enzyme involved in inflammatory signaling pathways. Preclinical studies demonstrated efficacy in animal models of asthma. Clinical development included several phase 1 trials evaluating safety, tolerability, pharmacokinetics, and formulation optimization in healthy volunteers and patients with mild asthma. Despite initial promise as an oral antiasthmatic agent, clinical development has been discontinued[1][2][3][5][9][10].
