BI-K0376 is a potent and selective small molecule inhibitor of the kinase **KNL1-binding protein** (KNL1-BP), also known as **Bub1 kinase** (Budding Uninhibited by Benzimidazoles 1). Developed by Boehringer Ingelheim, the compound is designed to disrupt the spindle assembly checkpoint (SAC) during mitosis. Bub1 kinase plays a critical role in chromosome alignment and the recruitment of other checkpoint proteins to the kinetochore. By inhibiting Bub1, BI-K0376 induces chromosomal instability and aneuploidy in rapidly dividing cancer cells, leading to mitotic catastrophe and apoptosis. It is primarily utilized as a high-quality chemical probe to study the biological functions of Bub1 in cell cycle regulation and as a lead candidate for potential antineoplastic applications.
