Bifunctional short hairpin RNA targeting furin (bi-shRNAi furin) is a specialized RNA interference (RNAi) molecule designed to knock down the expression of the proprotein convertase furin. Developed by Gradalis, Inc., this technology utilizes a proprietary "bifunctional" design that incorporates both a cleavage-dependent (siRNA-like) and a cleavage-independent (miRNA-like) component to enhance the potency and durability of gene silencing. In the context of cancer immunotherapy, specifically the autologous tumor cell vaccine gemogenovatucel-T (Vigil), bi-shRNAi furin is used to downregulate furin, which is responsible for the proteolytic activation of the immunosuppressive cytokines transforming growth factor beta 1 (TGFβ1) and beta 2 (TGFβ2). By reducing TGFβ levels within the tumor microenvironment, the vaccine aims to overcome local immune suppression and enhance the systemic anti-tumor response initiated by the co-expressed GM-CSF.
