Overview

bi-SNA (bimodal Spherical Nucleic Acid) is an experimental nanotherapeutic platform designed for the treatment of glioblastoma (GBM) and other high-grade gliomas. It consists of a 15-nm gold nanoparticle core functionalized with a dense shell of hairpin oligonucleotides that incorporate STAT3-decoy sequences. This bimodal design enables the simultaneous activation of the cyclic GMP-AMP synthase (cGAS) pathway to stimulate innate immunity and the sequestration of Signal Transducer and Activator of Transcription 3 (STAT3) to inhibit oncogenic signaling and immune suppression. To overcome the blood-brain barrier (BBB), bi-SNA is delivered via a novel facial dermal iontophoresis system that leverages facial neuro-lymphatic and neuronal pathways (such as the trigeminal and olfactory routes) for targeted brain delivery, bypassing systemic exposure.
