BiAb-103C is a novel bispecific antibody targeting the innate immune checkpoints CD47 and CD24, developed by Biotheus Inc. It is constructed using an anti-CD47 variable domain of heavy-chain antibody (vHH) and a fully human monoclonal antibody against CD24 on a human IgG1 scaffold. The vHH component is specifically selected for strong ligand-blocking activity with minimal binding to red blood cells (RBCs), addressing the common toxicity of hemagglutination associated with CD47-targeting agents. BiAb-103C blocks the CD47-SIRPα and CD24-Siglec-10 don't eat me signaling pathways, thereby enhancing macrophage-mediated phagocytosis and antibody-dependent cellular cytotoxicity (ADCC) against tumor cells. Preclinical studies in humanized mouse models have demonstrated significant antitumor efficacy and a favorable safety profile.
