Bicalutamide + ribociclib is a combination therapy being investigated for the treatment of androgen receptor-positive (AR+) triple-negative breast cancer (TNBC). This combination pairs bicalutamide, a nonsteroidal anti-androgen that blocks androgen receptors, with ribociclib, a CDK4/6 inhibitor. The rationale behind this combination is that CDK4/6 inhibition may enhance the activity of androgen inhibitors, as seen in previous prostate cancer cell lines. Preliminary studies suggest the combination has tolerable toxicity profiles and may provide improved clinical benefit compared to either agent alone.
