Overview

*Bifidobacterium animalis* subsp. *lactis* J12 is a probiotic strain being investigated for its therapeutic potential in managing atopic dermatitis (AD). It has been studied in two primary formulations: oral live cells (J12L) and topical fermented supernatants (J12S). The oral formulation exerts systemic effects via the gut-skin axis, enhancing gut microbiota alpha diversity and modulating the immune system to reduce epidermal thickness and mast cell infiltration. The topical formulation provides local anti-inflammatory effects and protects the skin barrier. Both delivery routes have demonstrated the ability to reduce pro-inflammatory cytokines such as IL-4 and IL-17 while increasing the proportion of splenic regulatory T cells (Tregs). In preclinical mouse models, J12 has shown superior efficacy compared to other probiotic strains, highlighting its versatile potential as a live biotherapeutic for inflammatory skin conditions.
