**biglycan siRNA** is a small interfering RNA (siRNA) therapeutic targeting the mRNA of **biglycan**, a small leucine-rich proteoglycan involved in extracellular matrix regulation, angiogenesis, and tumor progression. By specifically delivering siRNA against biglycan to tumor endothelial cells, notably using targeted liposomal nanodevices (such as RGD-MEND), this agent silences biglycan expression in the tumor vasculature. The primary mechanism is RNA interference-mediated degradation of biglycan mRNA, resulting in decreased biglycan protein levels. Preclinical studies in renal cell carcinoma models show that biglycan siRNA therapy inhibits angiogenesis, suppresses tumor growth, normalizes tumor vessel morphology, reduces vessel sprouting, and improves pericyte coverage. The therapy has been explored experimentally for antiangiogenic cancer treatment, particularly targeting tumor endothelial cells and the tumor microenvironment[2].
