BIIB015 is a humanized IgG1 monoclonal antibody-drug conjugate (ADC) developed for the treatment of solid tumors. It consists of an anti-Cripto (TDGF1) monoclonal antibody linked via a cleavable disulfide linker to DM4, a maytansinoid cytotoxic agent. The antibody component specifically binds to Teratocarcinoma-derived growth factor 1 (TDGF1/Cripto), a tumor-associated antigen highly expressed in various solid tumors but largely absent from normal tissues. Upon binding and internalization by Cripto-expressing tumor cells, the DM4 payload is released intracellularly where it binds tubulin and disrupts microtubule assembly/disassembly dynamics. This results in inhibition of cell division and growth through mitotic arrest and apoptosis induction. Preclinical studies demonstrated robust antitumor activity in xenograft models of breast, lung, colon, and testicular cancers[1][3][6]. Clinical development reached Phase 1 for relapsed or refractory solid tumors[5].
