Overview

BIIB021 + trastuzumab is a combination therapy studied for the treatment of advanced HER2-positive breast cancer. BIIB021 (also known as CNF2024) is an orally available small molecule inhibitor of heat shock protein 90 (Hsp90), a molecular chaperone involved in the stabilization and function of several oncogenic proteins. Trastuzumab is a monoclonal antibody that targets the human epidermal growth factor receptor 2 (HER2), which is overexpressed in some breast cancers. The combination aims to inhibit tumor growth by both blocking HER2 signaling and destabilizing multiple client proteins required for cancer cell survival. This combination was evaluated in phase 1 clinical trials to determine safety, pharmacokinetics, and pharmacodynamics in patients with advanced HER2-positive breast cancer[1][2][3].
