BIIB022 is a non-glycosylated human IgG4 monoclonal antibody that specifically targets the insulin-like growth factor 1 receptor (IGF-1R). It acts as an allosteric inhibitor of IGF-1R, blocking the receptor’s activation by its ligands IGF-1 and IGF-2. Unlike other anti-IGF1R antibodies, BIIB022 lacks effector functions due to its IgG4 backbone and non-glycosylated structure. The drug was developed for oncology indications where the IGF-1R pathway is implicated in tumor cell survival, proliferation, invasion, and metastasis. Clinical studies have shown that it can be safely administered intravenously at doses up to 30 mg/kg every three weeks in patients with advanced solid tumors[3][6][9].
