Overview

BIIB057 is a potent and selective small molecule inhibitor of spleen tyrosine kinase (Syk). It is orally bioavailable and has demonstrated high selectivity for Syk with an IC50 in the low nanomolar range. By inhibiting Syk, BIIB057 blocks B cell receptor-mediated signaling and activation in whole blood. This mechanism underlies its potential utility in treating autoimmune diseases such as rheumatoid arthritis and systemic lupus erythematosus, as well as certain cancers like non-Hodgkin lymphoma. The compound was originally developed by Portola Pharmaceuticals (as PRT062607) before being acquired by Biogen (hence the code name BIIB057). Clinical development reached up to phase 2 for rheumatoid arthritis but was discontinued[1][2][5][7].
