BIIB063 is a potent, high-affinity monoclonal antibody engineered to antagonize CD40. It is designed as an IgG4P molecule with reduced Fc effector function and avoids half-antibody formation in vivo. The drug inhibits the CD40 pathway, which plays a key role in immune activation by affecting germinal center formation, immunoglobulin class switching, T cell activation, and cytokine release—thereby suppressing inflammation and autoimmunity. Developed by Biogen for autoimmune diseases such as Sjogren's syndrome, its clinical development was discontinued due to preclinical safety findings and high immunogenicity observed during early trials[1][2][3][4].
