BIIB122 (also known as DNL151) is a selective, central nervous system-penetrant small molecule inhibitor of leucine-rich repeat kinase 2 (LRRK2). It is being developed as a potential disease-modifying therapy for Parkinson's disease, particularly in patients with LRRK2 mutations but also in those without such mutations. The drug works by inhibiting the kinase activity of LRRK2, which is implicated in lysosomal dysfunction and neurodegeneration associated with Parkinson's disease. By targeting LRRK2, BIIB122 aims to improve lysosomal function and slow the progression of Parkinson’s disease symptoms. The drug has demonstrated substantial peripheral and central inhibition of LRRK2 activity in clinical studies and is currently being evaluated in multiple phase 2 trials for early-stage Parkinson’s disease[1][3][6][7].
