Overview

BIK387 is a novel, low molecular weight, orally available small molecule inhibitor of the B-cell activating factor (BAFF) signaling pathway. It specifically targets the interaction between BAFF and its receptor, BR3 (BAFF-R), which is critical for B-cell survival and differentiation. Developed by Nobelpharma, BIK387 has demonstrated the ability to inhibit the differentiation of B cells into plasma blasts and plasma cells, suppress IgG production, and reduce the expression of key transcription factors such as AICDA, Blimp-1, and IRF4 in vitro. In preclinical models of systemic lupus erythematosus (SLE), oral administration of BIK387 significantly reduced renal dysfunction markers and decreased plasma cell proportions in the blood and spleen. It is being investigated as a potential therapeutic candidate for autoimmune diseases, including SLE and Sjogren's syndrome.
