biNTe (bispecific iNKT cell engager) is a novel therapeutic platform designed to recruit and activate invariant Natural Killer T (iNKT) cells against cancer cells. Developed by researchers at Imperial College London, the biNTe molecule features a bispecific architecture where one arm specifically binds to the invariant T cell receptor (iTCR) of iNKT cells, while the other arm targets a tumor-associated antigen, such as B-cell maturation antigen (BCMA). This engagement triggers iNKT cell-mediated cytotoxicity against tumor cells. Unlike conventional T-cell engagers, biNTe leverages the unique properties of iNKT cells, which bridge innate and adaptive immunity and can be used in an "off-the-shelf" allogeneic setting without the risk of graft-versus-host disease (GVHD). Preclinical studies have demonstrated potent activity against multiple myeloma cell lines and patient-derived plasma cells.
