bintrafusp alfa + cisplatin + paclitaxel is a combination therapy comprising bintrafusp alfa, a bifunctional fusion protein targeting both programmed death ligand 1 (PD-L1) and transforming growth factor beta (TGFβ); cisplatin, a platinum-based chemotherapy; and paclitaxel, a microtubule-stabilizing agent. Bintrafusp alfa functions as both an immune checkpoint inhibitor and a TGFβ "trap," enhancing antitumor immune responses and reversing immune resistance. Cisplatin and paclitaxel are cytotoxic chemotherapies commonly used in various solid tumors. This combination is being investigated mainly in the context of HPV-associated malignancies and other advanced cancers, aiming to exploit both immunotherapy and chemotherapy mechanisms for potentially synergistic effects, especially where TGFβ-mediated immunosuppression and chemoresistance are key issues[1].
