Bintrafusp alfa + paclitaxel is a combination therapy being investigated for various cancer types, particularly in recurrent/metastatic cancers. This combination pairs a novel bifunctional fusion protein (M7824, also known as bintrafusp alfa) with paclitaxel, a well-established chemotherapy agent. M7824 (bintrafusp alfa) is a first-in-class bifunctional fusion protein composed of an anti-PD-L1 monoclonal antibody fused to a TGF-β "trap". This innovative design allows it to simultaneously target two important immunosuppressive pathways in cancer: 1. It blocks PD-L1, preventing cancer cells from evading immune detection. 2. It sequesters TGF-β (transforming growth factor beta), which can promote tumor growth and suppress immune responses. The combination with paclitaxel, a cytotoxic chemotherapy that disrupts microtubule function during cell division, is being investigated to potentially enhance anti-tumor efficacy through complementary mechanisms of action.
