BIO-1819578 is a small molecule developed as a high-affinity inhibitor of *O*-GlcNAcase (OGA), an enzyme that removes O-GlcNAc modifications from proteins, and is radiolabeled with carbon-11 ([11C]) for use as a positron emission tomography (PET) imaging agent. Its main application is as a radioligand for non-invasive quantification of OGA levels and for measuring target occupancy of OGA inhibitors in the brain, which is relevant for neurodegenerative diseases such as Alzheimer's disease and other tauopathies. The tracer demonstrates specific, high-affinity binding to OGA, and it is suitable for reliable quantification using kinetic modeling in non-human primate models. PET imaging with [11C]BIO-1819578 can provide important information on brain OGA expression and assist in drug development targeting tau pathology by enabling studies of target engagement and dose selection for OGA inhibitors. The molecule was developed by Biogen[3][1][2].
