BIO 300 is a proprietary pharmaceutical formulation of synthetic genistein, an isoflavone that acts as a highly selective estrogen receptor beta (ERβ) agonist. Developed by Humanetics Corporation, it is designed as a radioprotective agent to prevent or mitigate acute radiation syndrome (ARS), delayed effects of acute radiation exposure (DEARE), and to reduce radiation-induced toxicities in cancer patients undergoing radiotherapy. Genistein’s mechanism involves modulation of anti-proliferative, anti-inflammatory, and anti-fibrotic gene expression via ERβ activation in hematopoietic cells, the gastrointestinal tract, and lungs. The drug has been formulated for multiple routes including oral powder/suspension and injectable suspension using nanoparticle technology to overcome poor oral bioavailability. Clinical development includes use as a medical countermeasure for military/civilian populations at risk of radiation exposure, protection against side effects from cancer radiotherapy (notably in non-small cell lung cancer), and mitigation of impaired pulmonary function following COVID-19 hospitalization[2][3][5][7][8].
