Overview

BioE-1115 is a small molecule inhibitor of Per-Arnt-Sim Kinase (PASK), a nutrient-sensitive protein kinase that regulates glucose and lipid metabolism. PASK is implicated in the progression of triple-negative breast cancer (TNBC) by promoting tumor growth through metabolic and signaling pathways. Preclinical research presented at AACR 2026 suggests that BioE-1115 exhibits differential efficacy across TNBC cell lines of distinct ancestral backgrounds, with African-ancestry cell lines showing reduced sensitivity compared to European-ancestry lines. Furthermore, the inhibitor's efficacy is modulated by glucose availability, highlighting the potential for BioE-1115 as a targeted metabolic therapy that requires consideration of both genetic ancestry and metabolic stressors.
