BioLexa

BioLexa is a topical combination therapy developed by Hoth Therapeutics for the treatment of atopic dermatitis. It utilizes a patented platform that combines a zinc-chelating agent, pentetic acid (DTPA), with the aminoglycoside antibiotic gentamicin. The mechanism of action involves disrupting the biofilm produced by *Staphylococcus aureus*, a bacterium that frequently colonizes the skin of patients with atopic dermatitis and contributes to disease flares and treatment resistance. By chelating zinc ions necessary for biofilm stability, the therapy breaks down the protective bacterial matrix, allowing the antibiotic to effectively clear the infection and reduce skin inflammation. BioLexa was licensed from the University of Cincinnati and is currently being evaluated in Phase 1 clinical trials for safety and efficacy in healthy subjects and patients with mild to moderate atopic dermatitis.
