BioLink2020 is a small molecule pan-FGFR inhibitor developed by Protheragen for the treatment of solid tumors. It specifically targets the fibroblast growth factor receptors 1, 2, and 3 (FGFR1-3), which are tyrosine kinase receptors that play critical roles in cell proliferation, survival, and angiogenesis. Dysregulation of FGFR signaling through mutations, gene amplifications, or chromosomal translocations is a known driver in various malignancies, including bladder, lung, and breast cancers. By inhibiting the kinase activity of FGFR1-3, BioLink2020 aims to block downstream oncogenic signaling pathways such as MAPK/ERK and PI3K/AKT. The compound is currently in Phase I clinical development as a targeted therapy for patients with advanced solid tumors harboring FGFR alterations.
