BiOncoFAP-11 is a high-affinity, bivalent small-molecule ligand designed to target Fibroblast Activation Protein (FAP), developed by Philogen. FAP is a serine protease that is highly overexpressed on the surface of cancer-associated fibroblasts (CAFs) in the stroma of more than 90% of epithelial tumors, while its expression in healthy adult tissues is negligible. BiOncoFAP-11 is a dimeric derivative of the OncoFAP ligand, engineered to enhance binding avidity and tumor retention through bivalent interaction with the target. It is primarily utilized as a targeting moiety for radiopharmaceutical applications, where it is conjugated to chelators such as DOTATATE for labeling with diagnostic radionuclides (e.g., Gallium-68) for PET imaging or therapeutic radionuclides (e.g., Lutetium-177) for radioligand therapy. The bivalent nature of BiOncoFAP-11 provides superior tumor-to-organ ratios and prolonged residence time compared to monovalent counterparts, making it a promising candidate for the precision treatment of various solid malignancies.
