BIOT-NFL-PEG-AuNPs is a **hybrid gold nanoparticle formulation** functionalized with biotinylated NFL-TBS.40-63 peptide and PEG (polyethylene glycol). The NFL-TBS.40-63 peptide specifically destabilizes the microtubule network within target cancer cells, originally demonstrated in glioma and further validated in pancreatic ductal adenocarcinoma (PDAC). Conjugation to gold nanoparticles and PEG enhances cellular uptake, targeting, and optimizes the peptide's anti-tumor effects. In preclinical models, BIOT-NFL-PEG-AuNPs slowed tumor growth, reduced tumor index, exhibited antiangiogenic effects, altered cancer cell metabolism, and modulated the immune response with minimal systemic toxicity. The primary mechanism is disruption of the microtubule cytoskeleton in cancer cells, leading to selective cytotoxicity against tumor cells, especially in PDAC[1][2][3][5].
